PT-141 Dosage Calculator: Vial Reconstitution and Units
PT-141 Reconstitution Calculator
Reconstituting PT-141
A PT-141 10mg vial with 2 mL of bacteriostatic water gives 5 mg per mL, so each unit on a U-100 syringe holds 50 mcg.
The same arithmetic applies to Melanotan II, supplied as a 10mg vial; choose it from the preset list.
PT-141 reconstitution chart
Concentration for each vial size and water volume. On a U-100 insulin syringe, 10 units is 0.1 mL and 100 units is 1 mL.
| Vial | Bac water added | Concentration | Per 10 units (0.1 mL) | Per 1 unit |
|---|---|---|---|---|
| 10 mg | 1 mL | 10 mg/mL | 1 mg (1000 mcg) | 100 mcg |
| 10 mg | 2 mL | 5 mg/mL | 0.5 mg (500 mcg) | 50 mcg |
| 10 mg | 3 mL | 3.333 mg/mL | 0.333 mg (333.333 mcg) | 33.333 mcg |
The formula behind the calculator
concentration (mg/mL) = PT-141 in vial (mg) ÷ water added (mL)
units on a U-100 syringe = amount (mg) ÷ concentration (mg/mL) × 100
Amounts in micrograms are divided by 1,000 first. The calculator does this arithmetic only; it does not recommend an amount, and PT-141 from AminoBlend is supplied for laboratory research use only.

PT-141 Calculator Explained
The Four PT-141 Results, Line by Line
The calculator divides the 10mg of PT-141 (bremelanotide) in the vial by the diluent volume to give mg/mL. Multiplying that by 100 shows how many micrograms sit in 0.1 mL, the first ten graduations of a U-100 syringe. It then divides your microgram target by the concentration to give millilitres, and multiplies by 100 for the unit reading.
500 mcg of PT-141 From the 10mg Vial
- 1 mL of bacteriostatic water: 10 mg/mL, 1,000 mcg per 0.1 mL, so 500 mcg is 0.05 mL, or 5 units.
- 2 mL: 5 mg/mL, 500 mcg per 0.1 mL, so 500 mcg is 0.1 mL, or 10 units.
- 3 mL: 3.333 mg/mL, about 333 mcg per 0.1 mL, so 500 mcg is 0.15 mL, or 15 units.
Smaller PT-141 Targets Need the Larger Water Volume
A 100 mcg PT-141 aliquot shows why the choice matters: it is 1 unit at 1 mL, 2 units at 2 mL and 3 units at 3 mL. Only the 3 mL option keeps it clear of a single graduation, and even then a 0.3 mL syringe or an intermediate dilution gives a more reliable reading than a 1 mL barrel.
Bremelanotide Molarity From Milligrams
At about 1,025 g/mol, the cyclic heptapeptide in a 5 mg/mL stock is close to 4.9 mM, and one micromolar works out at roughly 1.03 mcg per mL. That near one-to-one link between micromolar and mcg per mL makes PT-141 among the simpler compounds to convert for receptor binding or cell assays.
Errors That Creep Into PT-141 Calculations
- Entering the 10 mL volume of the PT-141 nasal spray as diluent; the spray arrives already in solution and is not reconstituted.
- Switching to the Melanotan II preset and forgetting to switch back, so results are logged under the wrong compound.
- Typing 0.5 as mcg instead of mg, which shrinks the answer a thousandfold.
- Reading 15 units as 1.5 mL rather than 0.15 mL.
PT-141 Calculator Questions
How much bac water for 10mg PT-141?
1 to 3 mL. 2 mL gives 5 mg per mL, so every unit on a U-100 syringe holds 50 mcg. More water makes small amounts easier to measure.
Does the calculator work for Melanotan II?
Yes. Choose Melanotan II 10mg from the preset list, or enter 10mg as the vial amount; the conversion is the same for any lyophilized peptide.
